Any "rule" on what name goes on the signature card comes from within; i.e. it's bank policy. My suggestion has always been that a bank should be willing to open an account in whatever name the customer has identification to support.
That's intended to acknowledge some flexibility; e.g.:
Nathan Bedford Forrest
Nathan "The Wizard" Forrest
Nathan B. Forrest
N.B. Forrest
Whether it's Jane or Jane Sue should not make a difference to you. You have her DOB and SSN and that should be enough to distinguish her from any other Jane Doe.
As far as a signature is concerned, the UCC provides some pretty clear guidance: It's whatever the customer wants it to be.
